In situ hybridization with radiolabeled antisense RNA probes corresponding to the coding region of the GABA(B1) receptor subunit, the GABA(A) receptor subunit alpha6, and the neurospecific enolase were used to localize and quantify their gene expression during morphological development from 22 weeks' gestation through 24 weeks' postconceptional age. Images were then processed in a semiquantitative manner with computer imaging techniques to evaluate spatial relationships of subunit expression with ultrastructural features of immature neurons and oligodendrocytes. Significant differences in subunit expression patterns between grey and white matter regions within each age group were discerned. Expression of GABA(B1) receptor subunit was specific to neurons and oligodendrocytes whereas expression of the GABA(A) receptor subunit alpha6 was diffusely distributed and may be associated with both neurons and oligodendrocytes in all brain regions. Neuro-specific enolase was expressed by neurons and oligodendrocytes in all brain regions. Ultrastructural analysis revealed that GABA(B1) receptor subunit expression was related to features of immature neurons including differentiation of cell bodies, axons, and dendritic arborization. Oligodendrocyte-specific distribution of the GABA(A) receptor subunit alpha6 was found in germinal matrix with a gradual decrease in expression of the alpha6 subunit with maturation of the oligodendrocyte. The neuro-specific enolase was diffusely distributed in the germinal matrix with a transient increase in expression during oligodendrocyte differentiation in germinal matrix and periventricular white matter. The findings demonstrate that temporal and spatial expression patterns of important genes involved in GABAergic neurotransmission are related to specific structural features of immature neurons and oligodendrocytes. The expression of GABA(B1) receptor subunit and neuro-specific enolase in differentiating neurons and oligodendrocytes suggests the existence of temporally specific roles for the GABA(B) receptor and enolase in phas 3e33713323
